What it means
Tweaking means acting mad suspicious, paranoid, jittery, or way too spun up over something small. People say it when somebody's jumping to wild conclusions, reading danger that isn't really there, or just bugging out harder than the moment calls for. It can be serious, but a lot of the time it's said half-joking when someone's overdoing it.

"Bro, you're tweaking if you think we're waking up at 5 a.m. on vacation just to stand in line for pancakes."
"Why are you checking his location three times in ten minutes? You're tweaking, relax."

Where it comes from
In street use, tweaking comes from tweak and got tied to twitchy, paranoid behavior, especially through drug slang around meth use, where somebody tweaking meant visibly strung out and skittish. From there it widened in everyday speech, so now people use it for anyone acting overly suspicious, frantic, or irrational, even when no drugs are involved.
